MT-2 (Melanotan 2 Acetate) is a synthetic melanocortin receptor agonist and an analog of the natural hormone 3$alpha$-Melanocyte-Stimulating Hormone (4$alpha$-MSH).5 It is a short peptide primarily known for its potent ability to induce skin tanning (melanogenesis), providing a “sunless tan.”6 It is a non-selective agonist of multiple melanocortin receptors (MC1R, MC3R, MC4R), which also gives it secondary effects, notably appetite suppression and the induction of spontaneous penile erections.

Description

Melanotan 2 (MT-2) is a cyclic heptapeptide developed to mimic the effects of 8$alpha$-MSH, a pituitary hormone.9 It is a research chemical that acts on the Melanocortin Receptor System, which regulates a wide range of physiological processes.

Key Mechanism of Action:

  • Tanning (MC1R Activation): MT-2 binds to the Melanocortin 1 Receptor (MC1R) on melanocytes (skin cells) and stimulates the production of the darker pigment, eumelanin.
  •  This results in skin darkening (tanning) without the need for extensive UV exposure.
  • Sexual Function (MC4R Activation): By binding to the Melanocortin 4 Receptor (MC4R) in the central nervous system (hypothalamus), MT-2 can cause an increase in libido and is a potent initiator of spontaneous, non-sexually stimulated penile erections (a property that led to the development of the related, but distinct, approved drug bremelanotide).
  • Appetite Suppression (MC3R/MC4R Activation): Its action on the hypothalamic MC3R and MC4R also mediates a reduction in food intake and overall appetite.

Major Concerns and Risks:

Due to its non-selective action and unregulated nature, MT-2 use is associated with several short- and long-term side effects, including: facial flushing, nausea, appetite reduction, darkening/creation of moles (nevi), and long-term concerns regarding potential increased risk of melanoma (skin cancer).
